The 26-year-old appeared to pull his hamstring early in Tuesday’s game.
Alexander-Arnold has experienced a slow start at Real Madrid, featuring in just two of their first four La Liga fixtures this season.
He is set to return to Anfield for the first time since his exit at the end of last season.
The clash between two top seeds is scheduled for November 4, with both sides aiming to secure automatic qualification for the knockout stages.
Just two minutes and 40 seconds had been played in Madrid when Alexander-Arnold played a simple pass before immediately clutching his left hamstring.
He sat down on the turf and signalled for medical assistance, but it quickly became clear he would not be able to continue in Los Blancos’ Champions League opener.
He was replaced by skipper Dani Carvajal, who had started Madrid’s 2-1 victory away to Real Sociedad at the weekend as Xabi Alonso’s side maintained their perfect start to the season.
Alexander-Arnold faces a race to be fit for November’s Anfield clash, a match set to be charged given his Liverpool exit.
“I think first and foremost, I want to say it’s not an easy decision and there’s a lot of thought and feeling that has gone into it,” the England international said back in May.
“I’ve been here 20 years now, loved every single minute of it, achieved all my dreams, achieved everything I’ve ever wanted to here.
“Giving everything day in, day out for 20 years, I’ve got to a point now where I feel like I need a change, a new challenge for me as a player and as a person.
“And I think now is the right time for me to do that.”
